Total RNAs prepared from these cells were used for gene expression analysis by real-time RT-PCR for WNT (a) and DACT3 (b). (c) Effect of WNT inhibitor XAV939 on mineralization of SHED by Alizarin Red S staining. SHED were cultured for 7 days in odontogenic induction media with or without 10 μM XAV939 and with or without HP by the medium height of 5 cm (H: 5cm). The Alizarin Red-positive areas were quantified using ImageJ. Scale bar, 150 μm. The data are representative of three independent experiments with similar results, and error bars indicate standard deviations. Statistical analysis was performed using analysis of variance (**p < 0.01).
