Figure 5

Post-response quiescence lacks some characteristics of sleep homeostasis. (a) Correlation of the durations of quiescent bouts (QBs) with the preceding movement bout (MB) duration in lethargus (black) and UV SIS (blue). Squares represent average QB duration following MBs in each MB duration bin, bars represent SEM. Bout duration was significantly correlated for lethargus (slope = 0.089, p = 0.01, R2 = 0.32), but not UV SIS (slope = −0.028, p = 0.69, R2 = 0.0090). n = 17 animals, three replicates for lethargus, n = 12 animals, two replicates for UV SIS. (b) Mechanosensory response (gray bars) and PRQ (red) of WT background animals 2–12 h after EGF overexpression. **Denotes p < 0.01, two-sided Wilcoxon signed rank test with Bonferroni correction for four comparisons, note that 10 s stimulus animals were less quiescent after stimulus. n = 48 animals, four replicates. (c) Photosensory response (gray bars) and PRQ (blue) of WT background animals 2–12 h after EGF overexpression. **Denotes p < 0.01 (two-sided Wilcoxon signed-rank test with Bonferroni correction for four comparisons, note 30 s stimulus animals were less quiescent after stimulus). n = 23, two replicates for 1, 3, and 10 s, n = 24, three replicates, for 30 s stimuli). (d) (left) Average quiescence of WT background animals 2–12 h after EGF overexpression after mechanostimulus (dashed line) when the stimulus interrupted a quiescent bout (blue) or an active bout (red). n = 28 animals, four replicates. These data also appear in Fig. 2. (right) Comparison of PRQ from the left. Data from 4-2 min before the stimulus were used for baseline quiescence. Dots represent single animal averages for stimuli that interrupted quiescent or active bouts. PRQ was not significantly different (p = 0.17, two-tailed, two sample t-test). (e) Average normalized activity (top) and quiescence (bottom) following a single 1 s mechanostimulus (left) and duplex (15 s separating a 3 s blue light stimulus and 1 s mechanostimulus) (right) stimulus in WT background animals 2–12 h after EGF overexpression. Red and blue dashed lines represent mechanostimuli and blue light stimuli, respectively. n = 47 animals, two replicates. These responses are quantified in panel f. (f) Mechanosensory response (gray bars, left) of WT background animals 2–12 h after EGF overexpression after single and duplex stimuli, and PRQ (red, right) of the same animals after single and duplex stimuli. *Denotes p < 0.05 (two sample t-test). ∆ quiescence was calculated as in Fig. 1a. Dots represent individual animal averages and error bars and shading represent ± SEM.
