HQ formation upstream of a T7 promoter involved joint participation of RNA and DNA. SBED-GMP was incorporated into RNA during transcription. RNA retained in an HQ was cross-linked with the DNA upon irradiation by UV light. Cross-linking sites were detected by primer extension that terminated at the cross-linking site (blue bar).
