U3 snoRNA knock-down impacts the articular chondrocyte’s phenotype. Non-OA chondrocytes (n = 5 donors) were transfected with a U3 snoRNA ASO (U3 ASO) or scrambled (SCR) ASO and cultured for 24 h. (A) Expression of U3 snoRNA; (B) COL2A1; (C) SOX9; (D) NKX3-2; (E) RUNX2; (F) MMP13 and (G) IL6 mRNA levels were determined by RT-qPCR analysis. Data from U3 ASO samples were calculated relative to controls transfected with the SCR ASO. Results were normalized to relative total DNA content in parallel wells. Statistical significance was determined using 2-tailed paired Student’s t-tests. Bars show the mean (± SD). *P < 0.05, **P < 0.01, ***P < 0.001 versus control conditions.
