General methodology. Knockin mice expressing fluorescent PSD95 and SAP102 were imaged with Spinning Disc Confocal Microscopy (SDM) and with FIB-SEM. SDM allows the acquisition of large field, 2D fluorescent images, while FIB-SEM is an electron microscopy technique with a resolution in the scale of nanometres that generates 3D stacks of images, but with a smaller field of view. We have used a combination of both techniques to estimate the actual densities of synapses per unit volume brain-wide.
