Caspase 3, caspase 8 and caspase 9 are not activated in Enterovirus A71-infected neuronal cells. Neurons derived from human NSCs were infected with EV-A71 at an MOI of 2. (A) Immunofluorescence images of active caspase 3 and Tuj1 in EV-A71-infected neurons at 12, 24 and 48 h postinfection. Green indicates active caspase 3, red indicates Tuj1, and DAPI was used to label nuclei. Scale bar represents 20 μm. (B) Neurons derived from human NSCs were infected with EV-A71 at an MOI of 2, and total protein was extracted at the indicated time points. Western blotting was performed to measure the expression of precleaved and active caspase 3 and EV-A71 3Dpol. (C, D) Neuronal cells derived from IMR-32 cells were infected with EV-A71 at an MOI of 2, and total protein was extracted at various time points. The expression of precleaved and active caspase 3, caspase 9 and caspase 8 was measured with western blotting. β-actin was used as an internal control in all western blots.
