Identification of USF2 as a transcription factor of BEX2. (A,B) Luciferase assay using 293 T cells transfected with pNL1.1-Nluc-Neo expressing the indicated BEX2 promoter regions. (C) A region of the BEX2 promoter sequence. Underlined sequence is the predicted binding site of USF2. (D) Luciferase assay of 293 T cells transfected with the indicated promoter region in pNL1.1-Nluc-Neo vector, and pcDNA4-hUSF2-myc or empty vector (EV). (E,F) Real-time PCR of HuCCT1 (E) and RBE (F) cells. The cells were knocked down using siRNA against human USF2. After 3 days, cells were harvested and total RNA was extracted. (G) Luciferase activity assay of pNL1.1-BEX2promoter2k-Nluc-Neo was transfected into 293 T cells that were starved for 24 h. pGL4.10[luc2] was co-transfected into 293 T cells as a control.
