CD20-transgenic T cells display normal growth properties. (A) FACS-analysis of CD20 + T cells. Primary T cells were isolated from peripheral blood and transduced with a CD20-encoding retroviral vector. The transduced cells were purified by immunomagnetic cell sorting based on CD20 expression. (B) 1.0 × 10^6 primary T cells were transduced with the CD20 or SFCMM-3 retroviral vector (mean of 50% CD20 + or 50% NGFR-positive) and the influence of CD20 expression on the viability of cells was investigated by Annexin-V and PI staining at 7 days post transduction. Early apoptotic cells were Annexin-V + / PI- and late apoptotic / necrotic cells were both Annexin-V + / PI + . (C) At day 7 post transduction the absolute numbers of viable cells were determined. The bars represent the mean ± SD of three independent transductions.
