Transfection of CGRP genes restored CGRP and the coding mRNA. (A) Profile of the in vitro experiments in dissociated cultured DRG neurons collected from the diabetic animals, at the end of the 8th week after induction of diabetes. The concentrations of glucose are labeled in the bracket under each group. Success of transfection of the vectors was primarily evaluated by visualization of green fluorescence in the neurons (B). Transfection of CGRP genes completely reversed the downregulations of CGRP coding mRNA (C) and CGRP (D). Control the neurons from the age matched non-diabetic animals, Diabetic the neurons from the diabetic animals, Db + LV-GFP DRG neurons from diabetic animals transfected with GFP genes, Db + LV-CGRP DRG neurons from diabetic animals transfected with CGRP genes. **p < 0.01, compared to the control; $$p < 0.01, compared to Diabetic group; &&p < 0.01, compared to Db + LV-GFP group.
