Detection of ZIKV in virus-spiked body fluid samples. Biological samples including urine, serum, saliva and semen were spiked with either a high (1 × 106 PFU/mL) or low viral load (1 × 103 PFU/mL) and processed for RT-LAMP without RNA extraction (A–I). The RT-LAMP results were observed by visual color change of the products in the reaction tube and by gel electrophoresis. Amplification products were observed by naked eye natural light (A, C, E and G), under UV irradiation (B, D, F and H) and agarose gel electrophoresis (I). Legends in (A–I) are: (1) NTC (non-template control): water; (2): uninfected biological sample (urine, serum, saliva and semen); (3): biological sample infected with 106 PFU/mL; (4): biological sample infected with 103 PFU/mL. M: molecular weight marker. Orange color: negative reaction for ZIKV; Greenish yellow color: positive reaction for ZIKV.
