Fidelity of DNA synthesis by saPolX. Primer extension assays were conducted to assess the ability of saPolX to incorporate dATP, dTTP, dGTP and dCTP opposite the four possible template nucleotides- dA, dT, dG and dC. The identity of the template nucleotide is mentioned at the top of each gel. wt-saPolX shows incorporation according to normal Watson–Crick base pairing (AD). H435A mutant shows accurate incorporation opposite templating bases A, G and C (EH) but exhibits significant misincorporation of dGTP opposite template dT (F).
