Effects of OLE on AQP2 phosphorylation and cAMP production in MCD4 cells. (A) Evaluation of cAMP production by FRET experiments in MCD4 cells transiently transfected with H96 as described in the “Materials and methods” section. Histogram reports netFRET measured in cells under basal conditions, treated with dDAVP (100 nM for 30 min), OLE (0.1 mg/ml O/N), or co-treated with OLE and dDAVP. The treatment with dDAVP displayed a significantly higher cAMP production depicted as a reduced netFRET signal. Data are shown as single values in a scatter plot reporting means ± S.E.M (***p < 0.001 vs CTR; $p < 0.05 vs dDAVP). (B) Lysates from MCD4 cells were subjected to immunoblotting using Abs against AQP2-pS256 and totalAQP2.
