Sensitivity of cell lines to MK-8776 and AZD6738. (A) Each cell line was incubated with MK-8776 (top) or AZD6738 (bottom) for either 24 h or 48 h, then drug was removed, and cells were incubated in fresh media for an additional 5–6 days. Alternately, cells were incubated in drug continuously for 6–7 days. Cells were lysed in the well, stained with Hoechst 33258, and the concentration that inhibited growth by 50% was recorded. Cell lines in both panels are sorted by sensitivity to MK-8776. The panel for MK-8776 is updated from our prior paper5. (B) Results for AZD6738 from A are resorted in order of sensitivity to AZD6738. The cell lines highlighted in red are MDA-MB-231 and its ATM-dysfunctional derivatives discussed later.
