Trilineage differentiation. This figure shows the differentiation of all 6 LND and all 6 control lines into the 3 primary germ cell layers via merged immunostains for ectoderm (PAX6 = green, nestin = red), mesoderm (Brachyury = green, NCAM = red), and endoderm (SOX17 = green, FOXA2 = red). All cultures were stained simultaneously, and all photomicrographs were taken using the same microscope settings. Distinct red or green colors are evident for ectoderm, where the markers were often expressed in different cells. Shades of orange and yellow result from varied proportions of overlapping red and green markers. Scale bar is shown at bottom right and depicts 100 μM.
