Engagement of PDGFRβ by TRAP distinguishes human and rodent infective Plasmodium species and is restricted at the level of TRAP. (A) PvTRAP ectodomain (ECD) staining reagent bound HEK293F cells transfected with human PDGFRβ (hPDGFRβ) in a concentration dependent manner that was not different from the binding of PfTRAP ECD. (B) PyTRAP ECD or a fragment containing the VWA + TSR domains did not bind to HEK293F cells transfected with mouse PDGFRβ (mPDGFRβ) at any concentration tested. (C) PfTRAP and PyTRAP ECD were tested at 1000 nM for binding to both hPDGFRβ and mPDGFRβ transfected HEK293F cells. PfTRAP ECD bound to both receptors, although less strongly to mPDGFRβ, while PyTRAP ECD did not specifically bind to either receptor. (D) Known ligand for hPDGFRβ and mPDGFRβ, human PDGF-BB bound to both receptors at 10 nM. Data are the mean ± SD from at least three independent experiments.
