Constructs of triple fluorescence flow-cytometry-based assay to analyze P-HES1 activity. Three plasmids are co-transfected: (I) activator plasmid with two open reading frames encoding the Notch intracellular domain (NICD) as the activator of the Notch pathway and the EGFP as transfection control for the activator plasmid. (II) the modulator plasmid, encoding the modulator protein of interest E6, which is N-terminally fused to mTagBFP2 to monitor E6 expression. (III) the reporter plasmid encoding for the reporter DsRed2. The expression of DsRed2 is under control of the HES1 promotor which is regulated by the Notch pathway.
