Seed PBMCs and LCLs

Add media to media chamber

Seed PBMCs and LCLs into cell chamber

Expansion

Collect and pool cells from each well

Wash each well

Seed cells (T-cells and LCLs) to each well (with or without IL-2)

Collect cells from cell chamber

Wash cell chamber

Seed cells (T-cells and LCLs) into cell chamber

Expand cell chamber

Remove 50% of media from media chamber

Add fresh media (with or without IL-2) to media chamber

Medium change

Remove 50% of media from each well

Add fresh media (with or without IL-2) to each well

Add IL-2 to media chamber

Harvest

Collect and pool cells from each well

Wash each well

Count cells

Collect cells from cell chamber

Wash cell chamber

Count cells
