Pre-treatment with Z-VAD(OMe)-FMK prevents CH-CM-induced caspase-3 and PARP activation and cell apoptosis in CCA cell lines. CCA cell lines were pretreated with Z-VAD(OMe)-FMK (pan-caspase inhibitor) for 1 h and subsequently treated with CH-CM (0%, 50% and 75%) for 24 h. (A) The protein expression levels of cleaved caspase-3 and cleaved PARP were analyzed by western blot. (B) The relative protein level analysis showed that expression of cleaved caspase-3 and cleaved PARP were significantly reduced following pre-treatment of Z-VAD(OMe)-FMK (p < 0.001). (C) Flow cytometric analysis with annexin V–PI staining showed reduced CCA cell apoptosis following treatment with Z-VAD(OMe)-FMK.
