Knockdown of claudin-3 in SVEC4-10 cells promotes cell migration. (a) SVEC4-10 cells were transfected with two independent shRNA plasmids to knockdown claudin-3 protein (shClaudin-3-1 & shClaudin-3-2) accompanied with the transfection of control plasmid (Control). RT-PCR analysis was used to examine the mRNA expression in the two knockdown cells compared to control cells. (***p < 0.001) (n = 3 per group) (b) Western blot analysis indicated the knockdown of claudin-3 protein in SVEC4-10 cell. (c) Representative images of tube formation of SVEC4-10 cells transfected with shClaudin-3-1, shClaudin-3-2, and control. Scale bar, 100 μm. (d) Number of junctions and total length were analyzed as indicators of tube formation ability via the ImageJ. (n.s. no significance, ***p < 0.001) (n = 3 per group) (e) Scratching assay was conducted and the photos were taken at 0 h and 16 h under a microscope. Scale bar, 100 μm. (f) Wound width was quantified and analyzed. (***p < 0.01) (n = 5 per group) (g) Transwell assay was conducted and the representative images were taken. Scale bar, 100 μm. Cells on the bottom side were counted and analyzed by t-test. (**p < 0.01) (n = 5 per group).
