Screening transcription factors potentially involved in CD271 expression. (A) Representative data of CD271 expression in HPCM2 cells. HPCM2 cells were sorted into a CD271-low fraction and cultured for 3 days, followed by determination of CD271 expression using flow cytometry. (B) Real-time PCR of CD271 mRNA. The sorted cells (day-0) and subsequently cultured cells (day-1, 2, and 3) were harvested and total RNA was purified. (C) Gene Set Enrichment Analysis. Comprehensive gene expressions were obtained using microarray and compared day-2, 3 versus 0, 1. Target genes of NFKB and RELA were upregulated in day-2, 3 cells. ES, enrichment score. (D) Luciferase assay. 293 T cells were transfected with vectors harboring the indicated genes and the CD271 promoter region containing a luciferase reporter. Luciferase activity was measured 2-days post-transfection.
