H2O2 increased primary cilium formation in HeLa cells. (A–D) HeLa cells were incubated with 50 μM H2O2 for up to 36 h (A, B). Cells were transfected with AccuTarget negative control-siRNA or TB4-siRNA for 24 h. Then, cells were incubated with 50 μM H2O2 (C, D). The cells were fixed and stained with antibody against Ac-tubulin. The representative image of primary cilia was observed with 400X magnification under fluorescence microscope. White arrows indicated primary cilia (A, C). The ciliated cells out of more than 1,000 cells in the absence (white) or presence (grey) of H2O2 were counted. PC frequency was evaluated by the blinded double scoring (B, D). Each result was the representative of experiments performed at least four times and each experiment was performed with four samples (n = 4) in the same group.
