From: Toxicity of extracellular alpha-synuclein is independent of intracellular alpha-synuclein

Effect of endogenous and exogenous alpha-synuclein (ASYN) on the Oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of mesencephalic neurons. OCR and ECAR measurements of WT and a-synuclein KO midbrain neurons with glucose as energy substrate challenged with mono- or oligomers of ASYN. OCR and ECAR analysis progresses from basal respiration, ATP- and proton leak-linked respiration after injection of oligomycin to maximal respiration after Carbonyl cyanide-4 (trifluoromethoxy) phenylhydrazone (FCCP) injection. Non-mitochondrial respiration resulting from RO injection has been subtracted from all values. (A) Analysis showed a genotype dependent decreased oxygen consumption rate throughout the experiment indicated by reduced OCR levels as well as (B) a genotype dependent decreased glycolytic activity. Treatments with mono- and oligomers did not have significant effects. Data were normalized to DNA content and n = 3 and 3 technical replicates were measured on each plate. Statistical analysis was done with n Prism 6.0 using a two-way Anova test.
