Effect of paraquat, rotenone or alpha-synuclein (ASYN) monomers/oligomers on different neuronal types. Cortical (A), dopaminergic (B) and enteric (C) neurons untreated (Control) or treated with paraquat (12.5 μM or 6.25 μM) or rotenone (10 nM). The numbers of NeuN+ neurons counted for each treatment were normalized to the Control. (A) In cortical neurons, pesticide exposure had a significant effect on neuronal survival (1-way ANOVA: F = 27.93; P < 0.0001). Dunnett’s multiple comparison’s test revealed significant neuronal loss compared to control in the paraquat 12.5 μM and rotenone 10 nM treated cultures (P < 0,0001) as well as in the paraquat 6.25 μM treated cultures (P < 0.01). (B) In dopaminergic neurons, pesticide exposure lead to a significant reduction of neuronal survival (1-way ANOVA: F = 15.86; P < 0.0001). Dunnett’s multiple comparison’s test revealed significant neuronal loss compared to control in paraquat 12.5 μM and rotenone 10 nM treated cultures (P < 0,0001) but not in the paraquat 6.25 μM ones (v> 0.05). (C) In enteric neurons, pesticide exposure significantly decreased neuronal survival (1-way ANOVA: F = 9.866; P < 0.0001). Dunnett’s multiple comparison’s test revealed significant neuronal loss in paraquat 25 μM (P < 0.001) and 12.5 μM (P < 0.05) but not rotenone 50 nM (P > 0.05) treated cultures compared to control cultures. Cortical (D), dopaminergic (E) and enteric (F) neurons untreated (Control) or treated with 1 μM or 0.1 μM ASYN oligomers, or 1 μM ASYN Monomers. (D) In cortical neurons ASYN monomer or oligomer exposure had no significant effect on neuronal survival (1-way ANOVA: F = 0.770; P = 0.516). (E) In dopaminergic neurons ASYN monomer or oligomer exposure had a significant effect on neuronal survival (1-way ANOVA: F = 32.45; P < 0.0001). Dunnett’s multiple comparison’s test revealed significant neuronal loss compared to control in the 1 μM monomers group (P < 0.001), as well as in the 0.1 μM (P < 0.001) and 1 μM (P < 0.0001) oligomers group. (F) In enteric neurons, exposure to monomers or oligomers did not have a significant effect on neuronal survival (1-way ANOVA: F = 0.6095; P = 0.6156). 6–8 embryos were used per experiment; each experiment was repeated at least 3 times.
