Intracellular cytokine staining. T cells were cultured under various conditions (as indicated) and stained for surface markers and cytokines (described in the materials and methods). The cells were first gated to exclude doublets or groups of cells, the live cells were selected by live/dead dye, followed by selection of CD3⁺ and CD4⁺ or CD8⁺. The cells could be further identified by cytokine expressions (the percentage of gated cells was indicated).
