Absence of ligase I does not alter the kinetics of DNMT1, UHRF1, G9A and GLP proteins on replicating DNA. The relative levels of; (a) DNMT1; (b) UHRF1; (c) G9A; and (d) GLP associated with EdU-labeled DNA in CH12F3 wild type cells (WT: L, blue line) and a LIG1 null derivative (LigI:H, red line) were determined by mass spectrometry as described in “Materials and methods”. (e) Immunoblotting of DNMT1, UHRF1 and histone H4 in chromatin fractions from CH12F3 wild type cells and a LIG1 null derivative. Data shown graphically are the mean ( ±) SEM of three independent experiments. Uncropped Western blots are shown in supplementary Fig S8.
