Proinsulin trafficking using RUSH in primary mouse β-cells. (A) Schematic of proCpepRUSH trafficking was created using biorender.com. (B) Mouse islets (C57BL6/J) treated with AdRIP-proCpepRUSH were examined 48 h post-infection. Biotin addition (200 μM) was used to initiate proCpepRUSH (green) trafficking. Islet cells were fixed at the indicated times post-biotin addition and immunostained for GM130 (magenta) and counterstained with DAPI (blue). (C) Specific time points from time-lapse imaging (Movie S1) of proCpepRUSH trafficking are shown with cell outlines.
