Procedure of culture preparation for further analyses (direct and indirect tests). For the direct tests, the PPmesh prostheses were incubated for 24 h in the complete medium. Afterwards, the cells were seeded (500 of 5 × 105 cells/mL) and incubated for another 24 h. Then, the prostheses were transferred to the new wells and incubated for either 1 or 4 days. For the indirect tests, prosthesis extracts were prepared through incubation of the prostheses in a complete culture medium for 24 h (at a volume equal to 1 mL per 0.1 g of the prosthesis). The extracts were frozen and thawed prior to analysis; cells cultured in 96-well plates were incubated with the extracts for either 1 or 4 days. In both the direct and indirect tests, further analyses were performed after 1 or 4 days of incubation using the cells or post-culture medium, depending on the assay.
