Translation is repressed during iron deficiency. U-2 OS cells were cultured for 2 days in the presence of the indicated concentrations of DFO, and treated with a 5-min pulse of puromycin at the indicated time points. Treatment with 200 μM sodium arsenite (NaAsO2) for 1 h was used as a control32. Proteins were extracted and analyzed by immunoblotting (see “Methods” section for details). Antibodies specific for puromycin and GAPDH were used. The Puromycin/GAPDH ratio is represented. The molecular weight (kDa) is shown on the right. (A) A representative experiment is shown.
