Analysis of respiratory quinones in aerobic and anaerobic culture conditions of strain PD-1 and reference strains. Cells cultivated in tryptic soy broth under aerobic and anaerobic conditions were used for extraction, purification, and quantification of the major ubiquinone 8 (Q8 [M-H]⁺ at m/z 728), demethylmenaquinone 8 (DMK8 [M-H]⁺ at m/z 704), and menaquinone 8 (MK8 [M-H]⁺ at m/z 718), detected by the reverse phase HPLC with UV detection at 254 nm and electrospray ionization-mass spectrometry in positive mode described in the Methods part and Supplementary Figure S5. As an exogeneous standard, ubiquinone 10 (Q10) was included.
