TbCDA-like colocalizes with stress granule marker proteins in T. brucei. (a) Colocalization of TbCDA-like with stress granules marker proteins TbDhh1, TbDRBD3 and TbScd6 under normal (control) and starvation conditions (120 min) in the TbCDA-like KI line with 6x c-myc fusion at the N-terminus. An anti-c-myc monoclonal antibody and the corresponding anti-Dhh1, anti-DRBD3 or anti-Scd6 polyclonal antibodies were used as primary antibodies. Alexa Fluor 488-conjugated anti-mouse IgG and Alexa Fluor 594-conjugated anti-rabbit IgG were used as secondary antibodies. Nuclei and kinetoplasts were labelled with DAPI. Images were acquired using a Leica DMi8 microscope and deconvoluted with Huygens Essential software (version 3.3; Scientific Volume Imaging). Bar, 10 μm. (b) Quantification of granules per cell with a minimum area of 0.05 μm2. Data represent the mean from at least 10 cells per condition, with boxes indicating the interquartile range and whiskers showing the maximum and minimum values.
