Replication stress affects the mitochondrial membrane potential and induces mitochondrial ROS production. (A) Induction of mitochondrial ROS by HU monitored by MITOSOX, in human SV40 transformed fibroblasts (RG37) and human osteosarcoma cells (U20S). (B) Induction of mitochondrial ROS by APH monitored by MITOSOX, in human SV40 transformed fibroblasts (RG37) and human osteosarcoma. (C) Analyse of the membrane potential alteration by HU or APH, in human SV40 transformed fibroblasts (RG37). (D) Mitochondrial ROS induction and mitochondrial membrane potential alteration by HU or APH in human primary fibroblasts. The data from at least three independent experiments are presented as the mean (± SEM) level of ROS production normalized to that of the control. The non-parametric Kruskal–Wallis test was used.
