USE-PCR accurately resolves and counts targets within complex mixtures. Three mixtures each containing a subset of synthetic tag templates were profiled on the QIAGEN QIAcuity (panels A, B, C; N = 6 replicates each) along with a mixture containing 31 synthetic tag tempates (panel All; N = 20 replicates). The signal from each partition was assigned to a corresponding tag identity and total counts for each tag are given on the y-axis (grey circles, log scale). Sequencing the same samples gave the corresponding NGS reads on the right y-axis (red diamonds).
