Frem2-KO mouse design strategy. A ES cells carrying the Frem2-KO-first, conditional-ready allele was used to generate the Floxed-Frem2 mouse line. The floxed allele shows the FRT-flanked Neo cassette (FLP and FRT system) inserted into exon 1, dividing it into 1a and 1b regions. The conditional-KO mouse harbors the following in-frame mutations (indicated as a red line): two insertions (27 bp and 30 bp) and one deletion (9 bp), with sequences provided below. B A portion of the sequence (15,100–15,120 bp) from exon 1 of Floxed-Frem2 is shown. The premature TGA stop codon sequence is highlighted in blue near the middle of the reading frame.
