HK-C60 produces IgA in human PBMCs. (A) IgA production in human PBMCs. PBMCs (1.0 × 107/mL, n = 6) were cultured with vehicle (PBS), anti-IgM (10 μg/mL) + CD40L (1 μg/mL), anti-IgM (10 μg/mL) + ODN2006 (5 μg/mL), HK-C60 (1.0 × 108, 2.5 × 108 or 5.0 × 108 CFU/mL) at 37 °C for 72 h. The IgA concentration in cultured medium was measured by ELISA. (B) In vitro cytokine neutralization assay. The PBMCs (1.0 × 107/mL, n = 6) were treated with vehicle (PBS) or HK-C60 (5.0 × 108 CFU/mL). The culture was further treated with anti-IL-6 and/or IL-10 mAb (both 10 μg/mL) or isotype antibody (10 μg/mL) at 37 °C for 72 h. The IgA concentration in the cultured medium was measured by ELISA. The data are shown as mean ± standard error mean (SEM) of six samples from two independent experiments. Student t-test or one-way ANOVA was used to analyze the data for significant differences. Values of *p < 0.05, **p < 0.01 and ***p < 0.001 were regarded as significant. ns: not significant.
