Assessment of CYP21A2 activity in human adrenal NCI-H295R cells. NCI-H295R cells were incubated for 4 h with various treatments: polymer, DMSO, ethanol, abiraterone, ABN (abiraterone nanoparticles), curcumin, piperine, PN (piperine nanoparticles), CN (curcumin nanoparticles), and CPN (curcumin piperine nanoparticles). The activity of CYP21A2 was determined using [3H]-17α-hydroxy progesterone as the substrate. Following incubation, steroids were extracted, dried, and separated by TLC. The conversion of 17α-hydroxy progesterone to 11-deoxycortisol was visualized through autoradiographic analysis on TLC plates and quantified using densitometry. One-way ANOVA was employed to statistically compare each treatment group to the control, with significance indicated as ****p < 0.0001.
