The GGGGCC (G4C2) repeat expansion in the C9orf72 gene is the most common cause of inherited amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). Three mechanisms are thought to contribute to ALS/FTD: loss-of-function due to decreased C9orf72 protein levels, RNA toxicity from the G4C2 repeats, and toxicity from dipeptide repeat proteins (DPR) produced via Repeat-Associated Non-ATG (RAN) translation.
