Real-time fluorescence imaging of newly acidified organelles during autophagy. a Hela cells were loaded with 1 (10 μM) for 30 min and were submitted to confocal microscope observation followed by addition of rapamycin (10 μM) under microscope. For each field, a series of 10–20 serial focal plane images was recorded, serial focal planes were 0.8 μm apart and were illuminated.
