Tag-seq profiling the off-target sites induced by AsCpf1 with 23 sgRNA in a single transfection. HEK293T cells were transfected with the donor Tag-oligo, AsCpf1 and a pool mix of 23 sgRNA. Cells were harvested three days after transfection, and genomic DNA was extracted for library construction and then sequenced. All the off-target cleavages of the 23 sites can be parallelly profiled by Tag-seq.
