TaPin1 (10 µM) was incubated for 24 h at 4 °C degrees with 500 µM DTM, MMV688754 or DMSO. Mixtures were diluted 1000-fold, and TaPin1 activity was assayed for 1 min by reading the absorbance at 380 nm. Apparent velocity constants (kobs) were extrapolated from these curves. While DTM effectively inhibited TaPin1 activity, MMV688754 did not have any inhibitory effect. These results are representative of three independent experiments, which are shown quantitatively below.
