The Ser/Thr kinase NEK6 colocalizes with LSD1 in specific chromatin sub-compartments (CSCs). The subsequent phosphorylation of LSD1 functions as a positive feedback loop leading to an increase in local substrate-enzyme-cofactor concentration and faster demethylation rates. This contributes to the establishment of multidimensional silencing hubs crucial for robust gene repression.
