Ssal-KRED hit variants were obtained via hot spot engineering (HSE), machine learning (ML) and iterative site mutagenesis (ISM). Positions are color-shadowed in accordance with the library they were investigated in (for reference, see Fig. 3ac). The fold-improvement over the wild type (FIOWT) was calculated from fold-improvement over the parent (FIOP) values. AA amino acid, − not applicable.
