Functional validation of Dendra2-labeled Tom40.

(A) Growth of wild-type (black) and Tom40-Dendra2 (red) was assayed on the YPGly non-fermentative media for 4 days at 30°C. (B) The rate of oxygen consumption by mitochondria isolated from wild-type (black) and Tom40-Dendra2 (red) strains was assayed polarographically using a Clark-type oxygen electrode (Hansatech Instruments). Oxygen consumption per mg of mitochondrial protein is obtained by normalization of the experimentally measured oxygen consumption rate by the concentration of the mitochondrial proteins determined by Bradford. The latter parameter varies between mitochondrial preparations.
