Fine-tuning Nodal/Activin/TGFβ signaling for efficient specification of paraxial mesoderm.

(A) Activin A/SB431542 dose-dependency. H9 and Mixl1-GFP hES cells and HK1 hiPS cells were differentiated in the presence of BIO, Noggin and various concentrations of Activin A or SB431542 and the MEOX1 and TCF15 transcripts were quantified by RT-PCR. (B) GSK3 inhibitor, Noggin and SB431542-dependent specification of paraxial mesoderm from Mixl1-GFP hES cells. RT-PCR was performed with MEOX1 and TCF15 primers. No, None; N, Noggin; AceBIO/Ace-BIO, Acetoxime-BIO; CHIR, CHIR99021; MeBIO, Methyl-BIO; S, SB431542. (C, D) Time-dependent expression of NODAL and BMP4. The H9 (C, D) and Mixl1-GFP (D) hES cells were differentiated with no factor (No), BIO or BIO + Noggin (BION) and subjected on the day indicated to RT-PCR.
