Conditional ko of the γ-pcdh locus.
(A) A viable conditional ko allele of the γ-pcdh gene cluster was generated by insertion of two LoxP sites on each side of constant region exon γ-C1. The schematic drawing shows the γ-pcdh locus with the 22 variable region exons (open boxes) and the three constant region exons (γ-C1 to C3, black boxes). The close-up below shows the wt allele and the targeted 3Lox allele containing the floxed lethal pgk-neo cassette. The floxed 2Lox allele containing two LoxP sites was obtained after excision of the pgk-neo cassette by crossings with EIIaCre mice. A functional γ-pcdh ko can subsequently be generated by Cre-mediated excision of “floxed” γ-C1. (B) Western blot analysis for γ-pcdh in wt, “floxed” (Lox/Lox) and γ-pcdh ko (ΔC1/ΔC1) mice. Quantification of western blots revealed no differences in γ-pcdh expression between wt and γ-pcdhlox/lox mice (relative signal intensity [%]), whereas in γ-pcdh ko mice no signal was detected (the asterisk marks unspecific bands).
