From: Induction of antiviral genes by the tumor microenvironment confers resistance to virotherapy
Acquired viral resistance is mediated by type I IFN in the conditioned media.
The effect of (a) anti-IFNα or (b) anti-IFNβ neutralizing antibodies on induction of luciferase expression in ISRE-Luc tumor cells and VSV mediated cell killing (MOI 1.0) of tumor cells that had been exposed 24 h to macrophage conditioned media or 10 U/ml IFNα or IFNβ. (c) IFN-receptor knockdown LM-1 cells (LM-1IFNARkd) are not responsive to the protective antiviral effects of RAW 264.7 macrophage conditioned media. Cells were exposed to the respective conditioned media and infected with VSV-GFP at indicated MOIs. Cell viability was determined 48 h later.
