Melting curves of Epac were determined in the presence of 100 μM cAMP. For ESI-09 and HJC0197 melting curves were determined at 1 μM, 2.5 μM, 10 μM, 25 μM, 50 μM and 100 μM and for ESI-05 at 0.1 μM, 0.15 μM, 1 μM, 2.5 μM, 10 μM and 25 μM. If two or more curves were highly overlapping the curves corresponding to the lower concentrations were omitted for clarity of presentation. The concentrations are indicated next to the curves in μM. Each melting curve was determined at least in duplicate.
