Procedure for SSC transplantation and offspring production.
Graft tissues were prepared from the testis of a fertile male (EGFP-expressing C57BL/6-Tg). The dissociated cells were cultured under the same SSC culture conditions. (B) In vitro cultured SSCs were microinjected into the seminiferous tubules of bulsufan-treated recipient mice (upper panel; 0.1% trypan blue-stained cells). Four weeks after transplantation, the recipient seminiferous tubules that had received EGFP-labeled donor SSCs and their cross sections were observed by fluorescence under UV light (middle and lower panels). Scale bars, 50 μm. (C) After ICSI, cultured blasotocysts emitted fluorescence under UV light. (D) An offspring derived from EGFP-labeled SSCs emitted fluorescence under UV light.
