NKG2D reverses the sULBP3-induced downregulation of NK cell cytotoxicity.
K562 (A), 7721 (B), or CD133−SW620 (C) cells were co-cultured with normal NK cells, with or without ULBP3-Fc (1 ng/ml), NKG2D-Fc (5 ng/ml) and NKp44-Fc (5 ng/ml). The cytotoxicity of NK cells was measured by LDH release after co-culture for 6 h at the indicated E:T ratios. (D) Normal NK cells were incubated with the indicated concentrations of ULBP3-Fc for 6 h and NKG2D expression in NK cells was measured by FCM. A representative graph of NKG2D expression is shown.
