Sensitivity analysis for PAGE-based genotyping approach.
(A) A total of 25 ng PCR template per reaction containing various ratios of wildtype and mutant clone DNA from the Agbl3 locus were tested in a 50 μl volume. Equal volumes of PCR products were loaded in each well of 2.5% agarose gel. (B) PCR products were analyzed using the T7E1 assay. (C) PCR products were further analyzed using PAGE-based approach. The minimal detection percentage for mosaic DNA templates in this assay was similar between T7E1 and PAGE-based approach, as shown in Figure 6B and 6C.
