Figure 4

miR-33b suppresses the migration and invasion of breast cancer cells in vitro.

(A,B) Transwell migration assays revealed that the ectopic expression of miR-33b inhibited the migration ability of BT-549 (A) and MDA-MB-231 (B) cells. (C,D) Matrigel-coated Transwell invasion assay revealed that the ectopic expression of miR-33b inhibited the invasion ability of BT-549 (C) and MDA-MB-231 (D) cells. (E) qRT-PCR analysis revealed that miR-33b downregulated the mRNA expression of metastasis-related genes LOX, MMP-2, MMP-9 and CXCR4 in BT-549 and MDA-MB-231 cells. (F) Western blot analysis of the metastasis-related proteins LOX, FN and p-FAK. miR-33b downregulated the levels of these proteins in BT-549 and MDA-MB-231 cells. The full-length blots were presented in the Supplementary Figure 9. Scale bars, 100 μm. Data represent mean ± s.d. *: P <0.05, **: P <0.01, ***: P <0.001.
